Seroconversion and seroreactivity patterns of dairy goats naturally exposed to caprine arthritis-encephalitis virus in Brazil Ciência Rural

A labelled avidin-biotin ELISA (lab-ELISA) using repeated serum samples of goats showed a progressive seroconversion with higher seroconversion rate at the period going from the beginning of the breeding up to the last half of lactation (35.0%), compared to that recorded at the beginning of breeding (17.8%)(p<0.05). Furthermore, the seroreactivity pattern, evaluated by a lab-ELISA standard-curve with serum samples collected at 30-40 days intervals during 12 months, was caracterized by high individual variability. RESUMO O objetivo do presente estudo foi investigar a soroprevalência de infecção com Toxoplasma gondii (T. gondii) em cabras em cinco províncias do noroeste da China. Soroprevalência foi determinado usando o kit de teste de ensaio de imunoabsorção enzimática (ELISA). A soroprevalência geral foi 21.23% (197/928). Análise estatística revlou que diferenças significativas foram observadas em fêmeas (P= 0.048, OR= 0.567, 95% CI= 0.309 a 1.041) e nos grupos ≥ 2 (P= 0.002, OR= 0.330, 95% CI= 0.224 a 0.488). The objective of this study was to compare the effect of hormonal treatment duration through the use of intravaginal sponges impregnated with 60 mg MAP for 6 (G6; n = 10), 9 (G9; n = 10) or 12 (G12; n = 10) days. All nulliparous (n = 15) and lactating (n = 15) Saanen goats received 37.5 µg d-cloprostenol and 200 IU eCG 24 hours before sponge removal during the nonbreeding season. The genetic diversity of Jamunapari goats (Capra hircus) was investigated using an optimized non-radioactive polymerase chain reaction single-strand conformation polymorphism (PCR-SSCP) method to detect <FONT FACE=Symbol>a</font>-lactalbumin polymorphism in a sample of 50 goats. Our data show that PCR-SSCP is an appropriate tool for evaluating genetic variability in Jamunapari goats. Polymorphism was detected in the sample, indicating that Jamunapari goats have high genetic variability at loci, exon I of the a-lactalbumin gene. This result opens interesting prospects for future selection programs and conservation strategies. Some Aspects of the Clinical Anatomy of the Mandibular and Maxillofacial Regions of the West African Dwarf Goat in Nigeria International Journal of Morphology

Some measurements in the mandibles and upper jaws of thirty West African Dwarf (WAD) goats without any apparent skeletal disorders were analysed. The length and height of the mandibles were 12.00 cm and 6.90 cm, respectively. The distance from the lateral alveolar root to the mental foramen, from the mental foramen to the caudal mandibular border were 1.56cm, 9.96cm, and 2.58cm respectively. The width and height of the mental foramen were 0.5 cm and 0.3 cm, respectively. These data are discussed with regard to their application to clinical manoeuvres around the head of the goat such as regional anesthesia during dental extraction

The objective of this study was to evaluate productive performance and milk quality of Saanen goats fed diets with soybean hulls replacing ground corn (0, 50 and 100% replacement) in early lactation (from birth to 50 days of lactation). Fourteen goats (72.30±9.14 kg of BW) were used in a completely randomized design. Animals were confined in individual stalls, with daily control of feed intake and milk production. On the 30th day of lactation, fecal sampling started in order to determine nutrients digestibility. This study describes the effect of sphingosine 1-phosphate (S1P) for development of preantral follicle, therefore the activation and follicular viability of caprine follicles cultured in vitro. Ovarian fragments were cultured for 1 or 7 days in Minimum Essential Medium with different S1P concentrations (0, 1, 10, 50, 100 or 200ng/mL). All ovarian fragments were processed for histological analysis in optical microscopy, transmission electron microscopy and fluorescence analysis. The treatment using 1ng/mL of S1P was able to maintain the percentage of normal follicles with the progression of the culture from day 1 to 7. Stable silencing of β-lactoglobulin (BLG) gene by lentivirus-mediated RNAi in goat fetal fibroblasts Genet. Mol. Biol.

β-lactoglobulin (BLG), a dominant allergen in goat milk, is difficult to remove by traditional biochemical methods. Its elimination from goat milk by genetic modification therefore poses a major challenge for modern goat breeders. A shRNA targeting BLG mRNA with high interference efficiency was identified, with which lentiviral vectors were used for mediating stable shRNA interference in goat-fetal fibroblast cells. Apart from high efficiency in the knockdown of BLG expression in these cells, lentivector-mediated RNAi manifested stable integration into the goat genome itself. Consequently, an in vitro model for goat BLG-content control was compiled, and a goat-cell line for accompanying transgenetic goat production created.

The genetic relationships and structure of fourteen goat (Capra hircus) populations were estimated based on genotyping data from 14 goat populations (n = 410 goats) at 13 microsatellite loci. We used analysis of molecular variance (AMOVA), principal component analysis (PCA) and F statistics (F IS, F IT and F ST) to evaluate the genetic diversity (Ho, He and ad) of the goats. Two active mutations (A 781 G and A 1575 G) in growth hormone (GH) gene, and their associations with litter size (LS), were investigated in both a high prolificacy (Matou, n = 182) and a low prolificacy breed (Boer, n = 352) by using the PCR-RFLP method. Superovulation experiments were designed in 57 dams, in order to evaluate the effect of different genotypes of the GH gene on superovulation response. The objectives of this study were to investigate whether TGF-β affect the survival, activation and further growth of goat primordial follicles enclosed in ovarian cortex after in vitro culture. Out of 1608 Nigerian Sahel male goats (bucks) examined for cryptorchidism in an abattoir, 9 (0.6%) had right unilateral cryptorchidism. The coat colour-specific prevalence was highest among the brown bucks (2.1%); and was 0.8%, 0.6%, and 0.3% among black, white, back-and-white bucks, respectively. The condition was not found among bucks with brown-and-black, brown-and-white, and multiple coat colours. Use of green fluorescent protein to monitor Lactobacillus plantarum in the gastrointestinal tract of goats BJM

The experiment aimed to specifically monitor the passage of lactobacilli in vivo after oral administration. The green fluorescent protein (GFP) gene was cloned downstream from the constitutive p32 promoter from L. lactis subsp. cremoris Wg2. The recombinant expression vector, pLEM415-gfp-p32, was electroporated into Lactobacillus plantarum (L. plantarum) isolated from goat. Green fluorescent protein (GFP) was successfully expressed in L. plantarum. After 2 h post-administration, transformed Lactobacillus could be detectable in all luminal contents. This study aimed at describing the distribution of vasa vasora in the tunica media of various parts of goat aorta, since this influences the physico-mechanical properties and disease occurrence. Specimens were obtained from ascending, arch, each vertebral level of descending thoracic and various segments of abdominal aorta of sixteen healthy adult male domestic goats (Capra hircus). The objective of the present study was to evaluate growth and development of corpus luteum (CL) using protein, DNA analysis and PCNA labeling index throughout the estrous cycle in goats. Non pregnant goats (n=20), 12 months of age were assigned for estrus synchronization. Goats CL were obtained from days 3, 8, 13 and 18 of estrous cycle using bilateral ovariectomy and blood samples were taken for plasma progesterone (P4) concentration.
